Dopamin, substantia nigra ile v\u00fccut hareketlerini kontrol eden di\u011fer beyin b\u00f6lgeleri aras\u0131nda mesajlar ileten bir kimyasald\u0131r. Dopamin insanlar\u0131n ak\u0131c\u0131 ve birbiri ile uyumlu hareketler yapmalar\u0131n\u0131 sa\u011flar. L-dopa kan beyin bariyerinden ge\u00e7er, AADC enzimi ile dekarboksilasyona u\u011frar ve dopamine d\u00f6n\u00fc\u015ft\u00fcr\u00fcl\u00fcr. B\u00f6ylece beyindeki dopamin eksikli\u011fi giderilmi\u015f olur. Ancak bu d\u00f6n\u00fc\u015f\u00fcm daha \u00e7ok konak\u00e7\u0131n\u0131n gastrointestinal sisteminde ger\u00e7ekle\u015fir ve L-dopa beyne gidemeden dopamine d\u00f6n\u00fc\u015fm\u00fc\u015f olur. bundan dolay\u0131 v\u00fccuda verilen L-dopa\u2019n\u0131n ancak %1 ile 5\u2019i beyne ula\u015fabilir. Periferde olu\u015fan dopamin ise kan beyin bariyerinden ge\u00e7emez. Ayr\u0131ca baz\u0131 yan etkileri bile olabilir. Vask\u00fcler dopamin resept\u00f6rleri arac\u0131l\u0131\u011f\u0131yla ortostatik hipotansiyona neden olabilir ve kardiak aritmileri art\u0131rabilir. Bunu \u00f6nlemek i\u00e7in Parkinson hastalar\u0131na L-dopa ile birlikte AADC inhibit\u00f6r\u00fc olan Carbidopa verilir. Ancak bu uygulamada bile L-dopa\u2019n\u0131n %56 s\u0131 beyne ula\u015famaz. <\/p>\n\n\n\n<p class=\"has-medium-font-size\">Ayr\u0131ca bir\u00e7ok \u00e7al\u0131\u015fmada n\u00f6rodejeneratif bozukluklar ile ba\u011f\u0131rsak bakterileri aras\u0131nda pozitif bir ili\u015fki saptanm\u0131\u015ft\u0131r. Yap\u0131lan bir deneyde Parkinson hastalar\u0131ndan al\u0131nan ba\u011f\u0131rsak bakterileri, bakterisiz fare modellerine nakledildi\u011finde farelerin motor eksikli\u011finin \u015fiddetlendi\u011fi g\u00f6zlenmi\u015ftir. Bu etki, antibiyotik tedavileriyle tersine \u00e7evriliyor. Tirozin Dekarboksilaz. Bu enzim ayn\u0131 anda hem L-dopa&#8217;y\u0131 hem de as\u0131l substrat\u0131 olan tirozini dekarboksile edebiliyor. AADC inhibit\u00f6r\u00fc olan Carbidopa&#8217;n\u0131n L-dopa&#8217;n\u0131n dekarboksilasyonunu tam olarak \u00f6nleyememesinin sebebi bu yolak. Yaln\u0131z bu enzim tirozine kar\u015f\u0131 L-dopa&#8217;ya g\u00f6re 5 kat daha fazla aktivite g\u00f6steriyor. Bilim insanlar\u0131 bundan dolay\u0131 tirozinin homologlar\u0131n\u0131 ara\u015ft\u0131rd\u0131 ve -a-florometiltirosin bile\u015fi\u011finin bu yola\u011f\u0131 inhibe edebilece\u011fini buldular. <\/p>\n\n\n\n<p class=\"has-medium-font-size\">Bu ke\u015fif parkinson hastalar\u0131na uygulanan L-dopa tedavisinin iyile\u015ftirilmesi a\u00e7\u0131s\u0131ndan b\u00fcy\u00fck \u00f6nem arz ediyor. Ara\u015ft\u0131rmac\u0131lar ke\u015fiflerinde daha ileri giderse parkinson hastalar\u0131 i\u00e7in belki de en \u00f6nemli tedavi \u015fekli olan L-dopa&#8217;dan \u00e7ok daha fazla verim al\u0131nabilecek ve beyne daha \u00e7ok L-dopa ula\u015fabilecek.
