Glioblastomalar\u0131n beynin d\u0131\u015f\u0131na yay\u0131lmas\u0131 son derece nadirdir. Sa\u011fl\u0131kl\u0131 h\u00fccrelerde telomeraz geni bu nedenle aktif de\u011fildir. \u00dcstel b\u00fcy\u00fcmeleri s\u0131ras\u0131nda, kanser h\u00fccrelerinde say\u0131s\u0131z mutasyon birikir.<\/p>\n\n\n\n<p style=\"font-size:22px\">Glioblastomlar erkeklerde kad\u0131nlardan biraz daha yayg\u0131nd\u0131r. Bilim insanlar\u0131 protein \u00e7evirisini \u00f6l\u00e7meye olanak sa\u011flayan modern ribozomal RNA dizilim teknikleri kullanarak beyin t\u00fcm\u00f6r\u00fc k\u00f6k h\u00fccrelerinin protein ekipman\u0131n\u0131 normal beyin k\u00f6k h\u00fccrelerininkiyle kar\u015f\u0131la\u015ft\u0131rd\u0131lar.<\/p>\n\n\n\n<p style=\"font-size:22px\">Kanser k\u00f6k h\u00fccrelerinde \u00fcretilen, ancak beyin k\u00f6k h\u00fccrelerinde \u00fcretilmeyen proteinler aras\u0131nda bir enerji metabolizmas\u0131 enziminin \u00f6zellikle ilgi \u00e7ekici oldu\u011funu g\u00f6rd\u00fcler. <\/p>\n\n\n\n<p style=\"font-size:26px\">Gliserol-3-fosfat dehidrogenaz 1 (GPD1) \u00fcreten kanser h\u00fccreleri b\u00f6l\u00fcnmedi ve esas olarak kanserin sa\u011fl\u0131kl\u0131 beyin dokusunu istila etti\u011fi yerler t\u00fcm\u00f6r\u00fcn s\u0131n\u0131rlar\u0131n\u0131 olu\u015fturdu. T\u00fcm\u00f6r geli\u015fiminin ba\u015flamas\u0131ndan yakla\u015f\u0131k iki hafta sonra, ara\u015ft\u0131rmac\u0131lar b\u00fcy\u00fcyen t\u00fcm\u00f6rde GPD1 \u00fcretimini tespit edebildiler.<\/p>\n\n\n\n<h3> <br>Gliserol-3-fosfat dehidrogenaz 1 (GPD1) <\/h3>\n\n\n\n<p style=\"font-size:22px\"> GPD1&#8217;in mayadaki strese duyarl\u0131 bir gen oldu\u011fu bilinmektedir. Ozmotik stres alt\u0131nda b\u00fcy\u00fcme i\u00e7in gerekli olan gliserol sentezinin temel enzimidir. Ekspresyonu, h\u00fccreler stres ko\u015fullar\u0131na maruz kald\u0131\u011f\u0131nda uyar\u0131l\u0131r. Bu gen NAD-ba\u011f\u0131ml\u0131 gliserol-3-fosfat dehidrojenaz ailesinin bir \u00fcyesini kodlar. Kodlanan protein, dihidroksiaseton fosfat\u0131n (DHAP) ve indirgenmi\u015f nikotin adenin din\u00fckleotitinin (NADH), gliserol-3-fosfata (G3P) ve NAD + &#8216;ya geri d\u00f6n\u00fc\u015f\u00fcml\u00fc d\u00f6n\u00fc\u015f\u00fcm\u00fcn\u00fc katalize ederek karbonhidrat ve lipid metabolizmas\u0131nda kritik bir rol oynamas\u0131na neden olur. Kanser k\u00f6k h\u00fccrelerinin stres sinyalleriyle sava\u015fmak i\u00e7in bu enzimi kulland\u0131\u011f\u0131 bulundu. Ayr\u0131ca GPD1 aktivitesine sahip h\u00fccrelerin kemoterapiye diren\u00e7li oldu\u011fu yap\u0131lan \u00e7al\u0131\u015fmalarla ispatland\u0131.<\/p>\n\n\n\n<p style=\"font-size:22px\">Ara\u015ft\u0131rmac\u0131lar standart kemoterap\u00f6tik ila\u00e7 olan Temozolomid\u2019i farelere uygulam\u0131\u015f ve tedavinin sona ermesinden sonra zaman i\u00e7inde farkl\u0131 noktalardaki t\u00fcm\u00f6rleri analiz etmi\u015flerdir. Tedavi s\u0131ras\u0131nda, GPD1 \u00fcreten kanser k\u00f6k h\u00fccreleri uyku hali durumunda kalmam\u0131\u015f n\u00fcks\u00fcn ba\u015flang\u0131c\u0131 ile birlikte, uykular\u0131ndan uyand\u0131klar\u0131n\u0131 g\u00f6zlemlemi\u015flerdir. Bu GPD1 \u00fcreten kanser k\u00f6k h\u00fccrelerin t\u00fcm\u00f6r\u00fcn tekrarlanmas\u0131ndan sorumlu olduklar\u0131n\u0131n g\u00fc\u00e7l\u00fc bir g\u00f6stergesidir. Y\u00fcksek GPD1 seviyeleri ayr\u0131ca renal h\u00fccreli karsinom gibi di\u011fer kanser t\u00fcrlerinde ilerleme ile de ili\u015fkilidir.<\/p>\n\n\n\n<p style=\"font-size:21px\"> GPD1; farelerin t\u00fcm\u00f6r k\u00f6k h\u00fccrelerinden CRISPR arac\u0131l\u0131\u011f\u0131yla genetik olarak elimine edildi\u011finde, hayvanlar\u0131n daha uzun s\u00fcre hayatta kald\u0131\u011f\u0131n\u0131 bulmu\u015flard\u0131r.<\/p>\n\n\n\n<p style=\"font-size:23px\" class=\"has-background has-very-light-gray-background-color\">CRISPR&#8217;I A\u00c7IKLADI\u011eIM YAZI:  <a rel=\"noreferrer noopener\" aria-label=\" (yeni sekmede a\u00e7\u0131l\u0131r)\" href=\"https:\/\/blog.ulubat.org\/index.php\/makale\/bir-arastirma-dosyasi-dunyanin-tasarlanmis-ilk-bebekleri\/\" target=\"_blank\">https:\/\/blog.ulubat.org\/index.php\/makale\/bir-arastirma-dosyasi-dunyanin-tasarlanmis-ilk-bebekleri\/<\/a><\/p>\n\n\n\n<p style=\"font-size:22px\">Farelerin t\u00fcm\u00f6rlerinde oldu\u011fu gibi, GPD1 \u00fcreten h\u00fccreler bask\u0131n olarak insan beyinindeki &nbsp;doku b\u00f6l\u00fcmlerinde istilac\u0131 olarak bulundu . Ara\u015ft\u0131rmac\u0131lar GPD-1&#8217;i genetik olarak kapatt\u0131ktan sonra glioblastoma h\u00fccreleri, beyin t\u00fcm\u00f6r\u00fc k\u00f6k h\u00fccrelerinin bir \u00f6zelli\u011fi olan t\u00fcm\u00f6r k\u00fcreleri olu\u015fturma yeteneklerini yitirdi\u011fi bulundu.<\/p>\n\n\n\n<blockquote class=\"wp-block-quote\"><p>T\u00fcm sonu\u00e7lar\u0131m\u0131z GPD1&#8217;in beyin t\u00fcm\u00f6r\u00fc k\u00f6k h\u00fccrelerinin istilac\u0131l\u0131\u011f\u0131ndan sorumlu oldu\u011funu g\u00f6stermektedir. Normal beyin k\u00f6k h\u00fccrelerinde, enzimin \u00f6zel bir rol oynad\u0131\u011f\u0131 g\u00f6r\u00fclmemektedir. Fareler GPD1 olmadan sorunsuz ya\u015fayabilir. GPD1 geni mutasyonlardan zarar g\u00f6rm\u00fc\u015f ki\u015filerde bile ciddi sa\u011fl\u0131k sorunlar\u0131 yoktur. Bu, ciddi yan etkileri tetiklemeden enzimi aktif maddelerle bloke etmenin m\u00fcmk\u00fcn oldu\u011funun \u00f6nemli bir g\u00f6stergesidir .
