Yan\u0131k besinler\nt\u00fcketme, UV \u0131\u015f\u0131nlar\u0131 ve y\u00fcksek s\u0131cakl\u0131k gibi etkenler bu genlerin mutasyona\nu\u011framas\u0131na neden olur. <\/p>\n\n\n\n<p>H\u00fccre\nd\u00f6ng\u00fcs\u00fcn\u00fcn veya \u00e7o\u011falmas\u0131n\u0131n d\u00fczenlenmesinde g\u00f6rev alan genler TP73, HDAC1,\nMSH2, IRS2, MLH1 ve TP63 gibi genlerdir. Bunlarda meydana gelen mutasyonlar\nh\u00fccrede sorunlar \u00e7\u0131kmas\u0131na sebep olur. \u00d6rne\u011fin MLH1 kolon kanserine, APC\nkolorektal kansere, NRG1 akci\u011fer kanserine ve BRCA2 meme kanserine yol a\u00e7ar.\nPeki bu mutasyonlar nas\u0131l meydana gelmektedir? <\/p>\n\n\n\n<p>DNA\nreplikasyonu sonucunda baz\u0131 hatalar olu\u015fabilir, \u00f6rne\u011fin bir n\u00fckleotide kar\u015f\u0131l\u0131k\ngelen n\u00fckleotid eksilebilir. Bunlar\u0131 onaran genler ATM, MSH2 ve MLH1\u2019dir.\nBeslenme d\u00fczensizli\u011fi bu genlerin protein sentezini azaltabilir ve dolay\u0131s\u0131yla\nDNA replikasyonu ger\u00e7ekle\u015firse yanl\u0131\u015flar\u0131n d\u00fczeltilmesi zorla\u015f\u0131r. Yine h\u00fccre d\u00f6ng\u00fcs\u00fcnde kademeli\nreaksiyonlarda g\u00f6revli IRS1 geni de h\u00fccre \u00e7o\u011falmas\u0131nda g\u00f6revli oldu\u011fu i\u00e7in bu\ngende meydana gelebilecek herhangi bir sorun kansere neden olabilir. Daha\nbir\u00e7ok genin de kansere neden oldu\u011fu bulunmu\u015ftur. <\/p>\n\n\n\n<p>Protein sentezleyen genlerde mutlaka sentezleme i\u015flemini aktive eden transkripsiyon fakt\u00f6r\u00fcn\u00fcn bulunmas\u0131 gibi inhibe eden fakt\u00f6r de bulunur. Bu sayede h\u00fccrenin s\u0131n\u0131rs\u0131z bir \u015fekilde o proteini sentezlemesi ve kansere d\u00f6n\u00fc\u015fmesi engellenir. mRNA\u2019n\u0131n post-transkripsiyonunda da kanser olu\u015fumunu engellemek i\u00e7in mekanizmalar bulunur. Ayn\u0131 \u015fekilde ba\u015fka k\u00fcfler de (<em>A. Parasiticus<\/em>, <em>penicilium,<\/em> <em>puberulum<\/em>&#8230;) Aflatoxin salg\u0131lar. Pirin\u00e7lere yerle\u015fen <em>P. Islandium<\/em> k\u00fcf\u00fc ise Luteoskyrin salg\u0131layarak karaci\u011fer kanserine yol a\u00e7ar. Bu da demektir ki pirin\u00e7 a\u011f\u0131rl\u0131kl\u0131 beslenen toplumlarda karaci\u011fer kanseri daha s\u0131k g\u00f6r\u00fclebilir.
