Young<\/em><\/p>\n\n\n\n<p><strong>\u00d6ncelikle sirkadiyen ritmik nedir? <\/strong><\/p>\n\n\n\n<p>Hayat\u0131m\u0131z\u0131n hep bir ritim i\u00e7inde ge\u00e7ti\u011fine &#8220;i\u00e7sel olarak&#8221; inan\u0131r ve ona g\u00f6re ya\u015famaya \u00e7al\u0131\u015f\u0131r\u0131z, \u00e7\u00fcnk\u00fc uyku zaman\u0131nda uyumam\u0131z\u0131, uyanmam\u0131z gerekti\u011finde uyanmam\u0131z\u0131 sa\u011flayan; tansiyon, v\u00fccut s\u0131cakl\u0131\u011f\u0131, kalp frekans\u0131 ve uyku-uyan\u0131kl\u0131k ritmi gibi unsurlar\u0131m\u0131z\u0131 y\u00f6neten sirkadiyen ritim diye adland\u0131rd\u0131\u011f\u0131m\u0131z biyolojik saatimiz bunu sa\u011flamaktad\u0131r. <\/p>\n\n\n\n<p>D\u00fcnya\n\u00fczerindeki t\u00fcm canl\u0131lar; bitkiler, hayvanlar ve insanlar ya\u015fam fonksiyonlar\u0131n\u0131\ng\u00fcne\u015fin hareketlerine g\u00f6re d\u00fczenlerler. <\/p>\n\n\n\n<p>K\u0131sacas\u0131 beynimizde \u00f6n&nbsp;<a href=\"https:\/\/sinirbilim.org\/hipotalamus\/\">hipotalamusta<\/a>&nbsp;yerle\u015fmi\u015f olan suprakiazmatik \u00e7ekirde\u011fin kontrol\u00fcndeki ritmimizdir. Beynimizin de dahil olmak \u00fczere neredeyse t\u00fcm organlar\u0131n\u0131z\u0131n kendilerine ait biyolojik ritimleri olup, i\u00e7 kaynakl\u0131 (endojen) bir uyar\u0131c\u0131 taraf\u0131ndan senkronize olurlar. <\/p>\n\n\n\n<p>Bu senkronizasyonun ba\u015f kahraman\u0131 suprakiazmatik \u00e7ekirdektir. Bu \u00e7ekirdek; biyolojik ritimlerin ana kontrol merkezidir ve hipotalamusta optik kiazman\u0131n \u00fczerinde yer al\u0131r<\/p>\n\n\n\n<p>Suprakiazmatik \u00e7ekirde\u011fin biyolojik ritmi d\u00fczenlemesindeki en \u00f6nemli d\u0131\u015f (ekzojen) kaynaktan biri \u2018\u2019\u0131\u015f\u0131k\u2019\u2019 etkenidir. G\u00fcne\u015f \u0131\u015f\u0131\u011f\u0131 almad\u0131\u011f\u0131nda bitkinin nas\u0131l bir davran\u0131\u015f sergileyece\u011fini merak ettiler ve onu karanl\u0131k bir ortamda beklettiler. Bilmedikleri bir gende\u00a0<a rel=\"noreferrer noopener\" href=\"https:\/\/www.drozdogan.com\/mutasyon-nedir-nasil-kanserlesmeye-neden-olur\/\" target=\"_blank\">mutasyon<\/a>\u00a0meydana getirmi\u015flerdi ve meyve sine\u011findeki biyolojik saatin mekanizmas\u0131 bozulmu\u015ftu. Bu geni\u00a0<strong>period<\/strong>\u00a0olarak adland\u0131rd\u0131lar fakat geni izole etmeyi ve etkisini tam olarak g\u00f6stermeyi ba\u015faramad\u0131lar. <\/p>\n\n\n\n<p> Nobel \u00d6d\u00fcl\u00fc&#8217;n\u00fcn iki ismi olan Jeffrey C. Hall ve Michael Rosbash mutasyona u\u011fram\u0131\u015f gen taraf\u0131ndan kodlanm\u0131\u015f olan bir protein (PER proteini) gece boyunca h\u00fccrede \u00fcretilerek depolan\u0131yordu. Fakat a\u00e7\u0131klanamayan baz\u0131 eksikler mevcuttu: bu  gece depolanan ve g\u00fcnd\u00fcz ayr\u0131\u015ft\u0131r\u0131ld\u0131\u011f\u0131n\u0131  nas\u0131l y\u00f6nlendiriyor? bu d\u00f6ng\u00fcy\u00fc nas\u0131l bir d\u00fczen i\u00e7erisinde oldu\u011fu bilinmiyor. Period geninin aktivitesini bloke etmek i\u00e7in,\nsitoplazmada \u00fcretilen PER proteini, gece boyunca genetik materyalin bulundu\u011fu\nh\u00fccre \u00e7ekirde\u011fine eri\u015fmek zorunda kalacakt\u0131r. <\/p>\n\n\n\n<p>Jeffrey Hall ve Michael Rosbash PER\nproteininin \u00e7ekirdekte gece boyunca biriktiklerini g\u00f6stermi\u015flerdi, fakat oraya\nnas\u0131l geliyordu? 1994 y\u0131l\u0131nda Michael Young, normal bir sirkadyen ritim i\u00e7in\ngerekli TIM proteinini kodlayan&nbsp;<em>\u201ctimeless\u201d<\/em>&nbsp;isimli\nikinci bir saat geni ke\u015ffetti. <\/p>\n\n\n\n<p>Bu&nbsp;<em>\u201ctimeless\u201d<\/em>&nbsp;geni TIM ad\u0131nda bir proteini \u00fcretiyordu ki bu protein sirkadiyen saatte olduk\u00e7a b\u00fcy\u00fck bir rol oynuyordu. Young, bu proteinin PER proteiniyle ba\u011flant\u0131l\u0131 \u00e7al\u0131\u015ft\u0131\u011f\u0131n\u0131, iki proteinin de h\u00fccre \u00e7ekirde\u011fine girebilmesini ve period geninin aktivitesinin durdurulmas\u0131n\u0131 sa\u011flad\u0131\u011f\u0131n\u0131 g\u00f6sterdi. <\/p>\n\n\n\n<p>Peki bu proteinlerin sal\u0131n\u0131m aral\u0131klar\u0131 nas\u0131l d\u00fczenleniyordu?
